📚 In-Depth Research Profile

Mechanism of Action

AHK-Cu is a synthetic copper-binding tripeptide consisting of alanine, histidine, and lysine (Ala-His-Lys) complexed with copper(II) ions. Structurally related to the naturally occurring GHK-Cu, its biological activity is heavily concentrated on endothelial cell regulation and follicular dynamics. It works primarily by modulating growth factors within the extracellular matrix, specifically upregulating Vascular Endothelial Growth Factor (VEGF) and Transforming Growth Factor-beta 1 (TGF-β1). By interacting with dermal papilla cells, AHK-Cu stimulates microcirculation and suppresses the pathways responsible for hair follicle regression, essentially protecting the follicular unit from stress-induced senescence (cell aging).

Therapeutic Uses

  • •Hair Growth Stimulation: Exhibits potent activity in promoting hair growth by expanding follicle size and significantly lengthening the anagen (growth) phase. It specifically targets and stimulates the proliferation of human dermal papilla cells.
  • •Anti-Alopecia / DHT Counteraction: Acts as a protective agent against androgenetic alopecia by counteracting the miniaturization of hair follicles, achieving this by reducing TGF-β1 secretion induced by dihydrotestosterone (DHT).
  • •Skin Rejuvenation & Vascular Support: Promotes the synthesis of collagen type I and elastin in fibroblasts. Its ability to stimulate endothelial cells enhances angiogenesis (the formation of new blood vessels), improving localized blood flow and skin elasticity.

Empirical Evidence

Pyo HK et al. (Arch Pharm Res 2007) — Demonstrated that Ala-His-Lys-Cu (AHK-Cu) significantly promotes the proliferation of cultured human dermal papilla cells and inhibits programmed cell death (apoptosis). The study confirmed that AHK-Cu stimulates hair elongation in isolated hair follicles, attributing its efficacy to the upregulation of VEGF and elongation of the anagen phase. Comparative in vitro assays indicate its follicular stimulation pathways are highly specialized, often showing higher cell-specific activity for dermal papilla cells than generic copper peptides.
