📚 In-Depth Research Profile
Mechanism of Action
5-Amino-1MQ is a cell-permeable, selective small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT consumes S-adenosyl methionine (SAM) and methylates nicotinamide, depleting both methyl-donor capacity and NAD+ precursors in adipose tissue. Inhibiting NNMT preserves SAM, restores NAD+ availability, and increases energy expenditure in adipocytes.
Therapeutic Uses
- •Adipose metabolic research: Reduced fat mass and improved insulin sensitivity in diet-induced obese mice without changes in food intake.
- •Muscle stem cell regeneration: Neelakantan et al. (2019) demonstrated improved muscle stem cell function and accelerated repair in aged mice.
- •Healthspan research: Of interest for sarcopenia and age-related metabolic decline.
Empirical Evidence
